Series stars and co-stars killed off (in the TV show)

Mark Giardina

Robert Lansing- 12 O'Clock High. Lansing's character (General Frank Savage) was Killed off in season two (because it was reported Lansing and producer Quinn Martin didn't get along and Lansing either left the show or was fired)

Charlie Sheen- Two and a Half Men. Need I say anymore.

James Garner- Nichols. Garner then played Nichols twin brother.

Henry Darrow- Harry O. His character, a police Lt., was killed off when the series moved to Santa Monica because of production costs. Anthony Zerbe was cast as Darrow's replacement.

Jean Stapleton- All in the Family. Actually I believe it was on Archie Bunker's Place that it was mentioned that Edith Bunker had passed away. Maybe I am wrong on that, but I remember the emotional scene as Archie found one of Edith's shoes under the bed and remembered her with fondness.

Diana Hyland on Eight Is Enough: She appeared as Joan Bradford in I believe 3 or 4 episodes of EIE in the first season before she died of cancer in 1977. The next season, Tom Bradford was a widower.

Kathleen Nolan on Real McCoys: Probably one of the biggest mistakes in sitcom history since when her character on the Real McCoys passed away and then when the show moved to CBS and then moved to Sundays opposite Bonanza on NBC. What once was a Top 10 show during the 1961-1962 season on ABC became a huge blood bath afterwards when they tried everything to save it even going so far as trying to get Luke to join up with a widow.

John Ritter - His death in real life resulted in the writers of his show having to feature a story about his character's demise.

Same for Redd Foxx who died on the set of The Royal Family.

I've got one - Martin Mull played Garth Gimble on Mary Hartman, Mary Hartman. Garth was a bad guy, and was impaled to death, if I remember correctly.

Producer Norman Lear wanted Mull as the host of the fake talk-show Fernwood Tonight. So Mull returned as Garth's twin brother Barth.

Jean Stapleton appeared in some episodes of "Archie Bunker's Place" during its first season, but chose to quit during that season, as she wanted to pursue other endeavors. At the start of the second season, it was revealed that Edith died of a stroke.
 
Freddie Prinze's suicide led to the demise of his character, Chico, on Chico and the Man.

Patrick Duffy left Dallas so Bobby Ewing was killed off. Then Duffy came back resulting in the infamous dream season which, for the following season, resulted in the 'deaths' of characters that didn't exist because the previous season didn't exist. But those characters didn't really die because, since it was a dream, they never existed anyway. ;D However, in the California world of Knots Landing, they acknowledged Bobby's death but didn't address his return so Bobby stayed dead there.

Also on Dallas, Jim Davis (Jock Ewing) died early in the series so Jock Ewing was killed off in a plane crash.
 
Sort of a minor character in this example, but the character of Josh Richland was killed off the original 90210, which led to Brandon becoming student body president at CU. Also, the way that they killed off Josh's character was not very believable, as his car exploded after what should have been a very minor fender-bender. ::)
