Due to the costs of the animation but it was one, if not perhaps the best ever, "serious" sitcom.

The Cape--DVD complete series release in 2011
The Courtship of Eddie's Father--First two seasons available on DVD
my beloved Everwood--All four seasons available on DVD
The FBI--Four (of nine) seasons released on DVD so far
Fish--Nothing so far
Marcus Welby--First two seasons (of seven) released on DVD so far
McCloud--I believe the first two seasons were released on DVD in 2005, but nothing since
Medical Center--First two seasons (of seven) released on DVD so far

On DVD: Maverick (first season so far), Wanted: Dead or Alive (all seasons), The Untouchables (all seasons)

On You Tube (some are presumably PD's): 30 eps of You Bet Your Life, 18(?) eps of S1 of Petticoat Junction, two eps of The Lucy Show (first season) entire first season of LA Law.

On Hulu: First three seasons of both Lou Grant and Hill Street Blues. Also on the site is the pilot for St. Elsewhere.

The Defenders is reportedly plagued by the producers' original contracts with the cast and crew that they would get higher royalties when they went into syndication. Thus, it wallows away, ignored in production company vaults, though one episode--chopped up into 4-5 segments can be seen on YouTube.

That was one of the best TV dramas of all time but it's difficult to believe old contracts are holding up a DVD release. Virtually all of the cast is deceased and most of the production crews must be as well.
 
I haven't seen "One Day at a Time" since its days on E!

Slight correction; it was supposedly concerning guest stars. Here is an item from six years ago on the Home Theater Forum website:

"I think I recall reading something to the effect that high residual payments for guest-stars rendered syndication fees for "The Defenders" (and perhaps "The Nurses" as well) too high, so they never circulated much in reruns. Don't know how true that is, or not."
 
Dallas ran in syndication for several years. Most recently, as I recall, on Soap.
 
The first two seasons of "Petticoat Junction" have been issued on DVD. I have them. It's the in-color seasons that are missing (except for bootlegs). I'm having to record them off of MeTV so I can get my fix of the hot babes ("daughters") who got hotter as the series progressed, and to follow the interesting way they did, or did not, deal with cast changes which included the dreaded death of two cast members.

"Amos and Andy" haven't been on TV, but there are more then one collection out on DVDs. The entire series of "Topper" hasn't been released as far as I can tell, just little packages with 4 or so episodes each.
 
When was the last time you saw an episode--even one episode--of Rowan and Martin's Laugh-In?

Sure, some of the jokes are going to be a little dated, but there's still a lot to laugh at.

I believe there are two DVD sets featuring a selection of episodes from around 1968 to 1971. The show's final 1972-1973 season was never released in syndication or on home video, as the Rowan and Martin-produced season were not up to par with the Schlatter / Friendly-produced seasons.
 
It shouldn't matter if one season of a long-running series was inferior in the view of whomever. Fans of the show would most likely want all seasons.
